General description

Phosphate buffer is the most commonly used component of irrigation solution during surgeries, to preserve the living tissues. It is also used in immunochemical staining procedures. Phosphate buffer serves as an important fixative for light and electron microscopy. Its buffering capacity is pH sensitive and is preferred to be used at physiological pH (7.2-7.4).

Application

Intended for use as a buffer in various Romanowsky type staining procedures, including Wright Stain, Wright Giemsa, Giemsa, May Grunwald, Jenner and Leishman. Phosphate buffer pH 7.2 at 25 °C has been used in oocyte collection from porcine ovaries.

Preparation Note

Each vial will prepare 3.8 L of buffer, 8.3 mM.

  2. could you provide me with the Wright-Giemsa protocol referenced in the question above about how to solubilize Product P3288, Phosphate Buffer pH 7.2. We need official instructions for use and the product did not come with it.

    1. If this product is ordered separately, no instructions are provided. Instructions for preparing P3288 are found in the WG Instructions for Use that ship with each bottle of WG16, WG32, WG80, or WG128. Below is the information from the Reagent section of the WG procedure Instructions For Use

      The phosphate buffer should be prepared by diluting 1 vial of buffer to 1 gallon or 3.8 liters of deionized water. Mix well to dissolve. Discard the working phosphate solution if turbidity or visible bacterial growth is present. Store the working phosphate buffer solution at 2-8°C

    1. The long term stability of this product in solution has not been evaluated. Upon reconstitution, the solution should be observed for any particulate matter or turbidity which would indicate possible microbial growth. However, similar prepared solutions such as P3619, a 1M Phosphate Buffer, are not assigned an expiration date. These products are quite stable and offer a warranty of 1 year from the date of shipment.

The instructions for reconstitution of Product P3288, Phosphate buffer pH 7.2 at 25 °C are indicated on the Wright-Giemsa protocol (see attached packaging insert).Briefly, Product P3288, Phosphate buffer pH 7.2 should be prepared by diluting 1 vial of buffer to 1 gallon or 3.8 liters of deionized water. Mix well to dissolve. Store Phosphate Buffer at room temperature (18-26°C). Store working phosphate buffer solution at 2-8°C. Discard the working phosphate solution if turbidity or visible bacterial growth is present.
