General description

Progesterone is a steroid hormone secreted by the ovaries.

Application

Use in cell culture applications to study sterol regulation of cell signaling and gene expression.
Progesterone has been used:
  • as a growth-promoting hormone in basal medium for bovine mammary epithelial cell culture
  • to test its neuroprotective abilities in a well-characterized lateral cortical contusion injury (CCI) model
  • as an endocytosis inhibitor to study its effect on Coxsackievirus A9 infection on A549 cells

Biochem/physiol Actions

Induces maturation and secretory activity of the uterine endothelium; suppresses ovulation.
Induces maturation and secretory activity of the uterine endothelium; suppresses ovulation. Progesterone is implicated in the etiology of breast cancer.
Progesterone is a hormone that maintains pregnancy. It is essential for embryo formation, development and survival in almost all mammals. This hormone exhibits its action via progesterone receptor, which is specifically expressed in the endometrial stroma throughout pregnancy. It is known to prevent preterm delivery, when administered intramuscularly. Progesterone also controls gonadotropin secretion, activates sexual behavior and triggers ovarian events leading to ovulation. It stimulates the growth and differentiation of the uterus and prevents uterine contractility.

Physical form

powder-RT; stock-frozen in working aliquots, avoid repeated freeze/thaw

Preparation Note

To prepare 20μg/ml stock solution, add 1ml absolute ethanol per mg progesterone, gently swirl to dissolve, add 49ml sterile medium per ml ethanol.

Other Notes

Steroid hormone produced by the corpus luteum.

Questions

1–4 of 4 Questions
  1. Why are some progesterone powders slighty yellow and others totally white? Wouldn't yellow indicate an impurity? Why is "slight yellow" allowed for identification on COA?

    1 answer
    1. This product is synthesized from materials derived from plant sources. Slight color variations from batch to batch are not out of the ordinary and are due to minor variations in the natural lower-level source material. The purity specification for the product is ≥ 99 % by HPLC; the batch-specific purity result would be listed on the corresponding CoA.

  2. What is the stability of progesterone reconstituted in ethanol and stored in the -80?

    1 answer
    1. The solution stability of this product at -80°C has not been investigated. Stock solutions prepared in ethanol are stable up to 6 months, when stored -20 deg C in the dark. Further stability studies have not been performed. Note that the Certified Reference Material, Product P-069, a 1 mg/mL solution of Progesterone in Acetonitrile, is stored at -20°C and assigned a shelf life of approximately 5 years.

      Helpful?

  3. How can a 10mg/ml stock solution of PHR1142 be prepared for injection into a mouse?

    1 answer
    1. The solubility of Progesterone in water is very low. For in vivo studies, the solubility of PROGESTERONE is 0.02 mg/ml in water and 39 mg/ml in H5784 (45% w/v 2-Hydroxypropyl-β-cyclodextrin). We do not run any in vivo studies with any of our chemicals. To use in animal studies, it is recommended to use the Cell culture tested product and also screen lots. P6149 and P8783 are the cell culture tested product numbers, and PHR1142 is a reference standard.

  4. What is the appropriate method for reconstituting item P8783, Progesterone?

    1 answer
    1. Progesterone dissolves in DMSO to 100 mM and in ethanol to 100 mM.
