Specificity

Recognition Sequence: 5′-AG/CT-3′
Ligation and recutting results: After 2-10-fold Alu I overdigestion of 1 μg λ DNA substrate, results in 95% cutting, >80% of fragments can be ligated, and >95% recut.
Heat inactivation: Inactivated at 65 °C for 15 minutes.

Application

AluI is restriction endonuclease used in molecular biology methods to cleave DNA at the recognition site 5′-AG/CT-3′, generating fragments with blunt ends.

Supplied with 10x Restriction Enzyme Buffer SA (B 7531).

Unit Definition

One unit is the enzyme activity that completely cleaves 1 mg of λDNA in 1 hr, at 37 °C in a total volume of 25 ml of 1x digestion buffer SA for restriction endonucleases.
