Streptokinase is an extracellular enzyme produced by β-hemolytic Streptococcus. It is a single-chain polypeptide enzyme that consists of multiple structural domains, including α-, β-, and γ-domains.

Application

Streptokinase has been used in a study to compare primary coronary intervention and thrombolytic therapy in myocardial infarction patients.
Streptokinase is commonly used as a thrombolytic agent in the therapy of ischemic stroke. This therapy carries the important risk of intracerebral hemorrhage. Streptokinase is also used in the treatment of complicated parapneumonic effusions and empyema where adverse reactions, allergic type, are rare.

Biochem/physiol Actions

Streptokinase is a potent plasminogen activator. Unlike human proteins tissue plasminogen activator (tPA) and urokinase plasminogen activator (uPA), microbial streptokinase does not directly activate plasminogen. Instead, it activates plasminogen by fibrin-dependent and fibrin-independent. mechanisms. Streptokinase is effective in thrombolytic therapy. While streptokinase offers promising potential in treating ischemic stroke, its use also presents a significant risk of intracerebral hemorrhage. Streptokinase is also used in treating complicated parapneumonic effusions and empyema where allergic reactions, are rare.

Physical form

Lyophilized powder containing ~50% total protein by biuret and sodium glutamate. Total protein composed of enzyme protein and human serum albumin.

    1. This product is lyophilized from a solution containing 11 mg sodium glutamate and 13 mg human albumin per 1 million units. The material is soluble in water, as well as in aqueous buffers compatible with the specific application. For testing purposes, the product is reconstituted in 10 mM Tris-HCl / 0.1 M NaCl / 0.1% BSA, pH 7.4. Working concentrations typically range from 5 - 20 units per mL and stock solutions may be prepared up to 1000 units per mL. Note that this enzyme is reversibly inactivated at or below pH 5 and irreversibly denatured at pH 9 or above. Prepared solutions are stable for several hours when refrigerated. For long term storage solutions may be stored at -20°C. Avoid repleated freeze/thaw cycles.
