General description

Thrombin has procoagulant and anticoagulant properties. It also has chemotactic and mitogenic effects. Thrombin triggers the growth of mammalian cells, fibroblasts and macrophage- like tumor cell lines. It regulates brain development.[1] Thrombin plays an important role in hemostasis and thrombosis. It is associated with mast cell degranulation along with the release of histamine and enhances the production of IL-1 (interleukin-1) by activated macrophages. Thrombin stimulates vascular permeability and transendothelial cell migration by neutrophils.[2]

Application

Thrombin is a serine protease involved in the coagulation cascade wherein it converts fibrinogen into fibrin. Thrombin may be detected (assayed) using peptides that contain the VPR sequence such as the fluorogenic substrates Boc-VPR-AFC, Boc-Val-Pro-Arg-7-AMC and the chromogenic substrate Boc-Val-Pro-Arg-pNA. These substrates may also be useful to detect other serine proteases.

    1. The fluorescence of a thrombin substrate, fluorogenic, typically increases upon cleavage by thrombin. These substrates are designed with a quenched fluorescence system, where the intact substrate exhibits low or no fluorescence. When thrombin cleaves the substrate at the specific recognition site, the fluorophore is separated from the quencher, resulting in an increased fluorescence signal.

    1. As per the supplier the product is soluble at 3 mg per ml or greater.

  3. How do we use this product i.e., its dissolution medium and its fluorescent intensity wavelengths (excitation and emission)?

    1. This material is used in research applications for testing mammalian cells as described in the general properties section of the product page. As per the supplier, the material can be solubilized in DMSO (> 3mg/ml) before use. The stock solution can be further diluted into buffer (e.g PBS). Stability of stock solutions is unrecorded. The Fluorophore is a Coumarin with an excitation wavelength at 365 nm and an emission wavelength at 440 nm (in ethanol(lit.)).
