Application

Zoniporide hydrochloride hydrate has been used for the inhibition of sodium-hydrogen exchanger isoform 1 (NHE-1) in rat models.[1] It has also been used in cytotoxicity assay and to study its effect on lymphocytic choriomeningitis virus multiplication.[2]
Zoniporide hydrochloride hydrate may be used in Na+/H+-mediated cell signaling studies.

Biochem/physiol Actions

Potent and selective inhibitor of Na+/H+ exchanger isoform 1 (NHE-1)
Zoniporide is a potent and selective inhibitor of Na+/H+ exchanger isoform 1 (NHE-1). IC50 = 59 nM at NHE-1, vs. 12,000 nM at NHE-2; Zoniporide inhibits NHE-1-dependent Na+ update with IC50 of 14 nM.
Zoniporide, a selective inhibitor of sodium-hydrogen exchanger isoform 1 (NHE-1) has cardioprotective effects against myocardial injuries and ischemic insults.[3] It inhibits the swelling human platelets and attenuates cardiac contractile dysfunction in rats.[4] Zoniporide might have neurotoxic effects as it causes peripheral sensory axonopathy.[5]
