General description

3,3′,5′-Triiodo-L-thyronine (T3) is an endocrine hormone produced by the thyroid gland and is regarded as an active thyroid hormone at cellular level. Thyroxine (T4) serves as a precursor for T3 and is generated in extrathyroidal tissues.

Application

3,3′,5-Triiodo-L-thyronine sodium salt has been used for inducing thyroid hormone manipulations in mice. It has also been used In cell culturing of hippocampal, retinal,[1] proximal tubule and glomerular explant.

Biochem/physiol Actions

3,3′,5′-Triiodo-L-thyronine (T3) increases the rates of protein synthesis, stimulates the breakdown of cholesterol and effects embryonic development. In cell culture, T3 regulates cell differentiation and protein expression. The interaction of T3 with nonhistone proteins in the chromatin initiates the thyroid hormone activity. Thyroid hormones regulate the differentiation and development of tissues and affect a number of metabolic processes. The level of circulating T3 hormone is reduced in hypothyroidism.

Preparation Note

To prepare 20 μg/ml stock solution: add 1ml 1N NaOH per mg 3,3′, 5-triiodo-L-thyronine; gently swirl to dissolve. To this, add 49 ml sterile medium per ml 1N NaOH added.

  2. I work with mammalian cell culture and using a protocol that calls for this product. I'm only aware of use of water, PBS, or DMSO to reconstitute cell culture media supplements. I've never seen use of NaOH. Just want to ask if it is safe to use NaOH here.

    1. There is nothing intrinsically harmful about NaOH, as it is can be used to properly pH cell culture media. When prepared in the recommended manner, the NaOH should have not appreciably affect buffered cell culture media.

    1. The stability of this product in solution at -80°C has not been determined. Stock solutions of this material may be stored in working aliquots at -20°C or at 2-8 °C for up to 30 days.
