General description

For spectrophotometric measurement of viable cells. Absorbance of converted dye is measured at a wavelength of 540 nm.

Application

In Vitro Toxicology Assay Kit, Neutral Red based has been used:
  • to measure the lysosomal enzyme activity in HB8902 cells[1]
  • to study the cell viability of human alveolar epithelial cells[2]
  • to study the cell viability in fibroblasts[3]

Biochem/physiol Actions

Neutral red (NR) is a red dye that accumulates in the lysosome by actively transferring across the cell membranes.[3] The dye is extracted and the uptake is quantitated by spectroscopy. The NR uptake assay is the most commonly used cytotoxicity test. It is widely applicable in environmental and biomedical studies to estimate the number of viable cells in culture. The NR dye in vitro tissue culture studies is used in assessing viral cytopathogenicity and detection of toxic compounds.[4]

  1. Can I use this kit for phototoxicity assay. If so, does the kit contain a positive control for phototoxicity?

    1. This kit utilizes Neutral Red, a vital dye, to measure cell viability after exposure to any specific potential toxin. Neutral red is only taken up by viable cells. Cell viability is determined by measuring the absorbance following the specified incubation period to the potential toxin of interest. A positive control is not included and would be the untreated cells. In this case, the viability of cells exposed to a given wavelength of light compared to cells that have not been exposed.
