Clone 4E15 is a Rabbit recombinant monoclonal antibody that specifically detects Rab7A. It targets an epitope within 17 amino acids from the C-terminal region.

Quality Control Testing
Evaluated by Immunohistochemistry (Paraffin) in human kidney tissue sections.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected Rab7A in human kidney tissue sections.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Rab7A in Cos-7 cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound Rab7A with a KD of 2.2 x 10-8 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Rab7A in HeLa, A431, HepG2 and NIH 3T3 cells.

Ras-related protein Rab-7A (UniProt: P51149; also known as Rab-7a) is encoded by the RAB7A (also known as RAB7) gene (Gene ID: 7879) in human. Rab7A is a highly conserved member of the large superfamily of Ras-like GTPases. It is widely expressed, peripheral membrane protein that is localized mainly to late endosomes and regulates transport from early endosomes to late endosomes and from late endosomes to lysosomes. It is not only important for vesicle formation, transport and fusion but also for cargo selection and sorting. Like other GTPases, Rab7 also undergoes characteristic cyclical mechanism of activation and inactivation depending on GTP binding and hydrolysis. It also plays a many other cellular and physiological events, such as growth-factor-mediated cell signaling, nutrient-transporter mediated nutrient uptake, neurotrophin transport in the axons of neurons and lipid metabolism. It also participates in some specialized endosomal membrane trafficking, such as maturation of melanosomes, pathogen-induced phagosomes and autophagosomes. More recently, it is reported to be essential for vesicular trafficking and cell surface expression of ACE2 and loss of Rab7A is reduces SARS-CoV-2 entry into cells by sequestering the ACE2 receptor inside cells through altered endosomal trafficking. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
