Calnexin (UniProt: P27824; also known as IP90, Major histocompatibility complex class I antigen-binding protein p88, p90) is encoded by the CANX gene (Gene ID: 821) in human. Calnexin is a member of the calreticulin family of proteins. It is a well-conserved, single-pass type I membrane protein that serves as calcium binding, endoplasmic reticulum (ER) resident chaperone protein. It associates with newly synthesized, incompletely folded monomeric glycoproteins and promotes their correct folding prior to secretion. It is also reported to target misfolded proteins for degradation. It is anchored to the ER membrane by a transmembrane domain. Calnexin associates with partial T-cell antigen receptor complexes that escape the ER of immature thymocytes and may, therefore, function as a signaling complex regulating thymocyte maturation. Calnexin is synthesized with a signal peptide of 20 amino acids, which is subsequently cleaved off to generate the mature form that has a lumenal domain (aa 21-481), a short transmembrane domain (aa 482-502) and a cytoplasmic domain (aa 503-592). Three isoforms of calnexin have been described that are generated by alternative splicing. Calnexin is shown to be phosphorylated at serine 564 by MAPK3/ERK1, which increases its association with ribosomes. Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
