Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Substance P in Jurkat cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Substance P in human cerebral cortex tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Substance P in SH-SY5Y cells.
Affinity Binding Assay: A representative lot of this antibody bound Substance P with a KD of 5.1 x 10-7 in an affinity binding assay.

Target description
Substance P, a member of the tachykinin family, is an 11 amino acid peptide that is derived from the Protachykinin-1 (UniProt: P20366) that is encoded by TAC1 gene (Gene ID: 6863) in human. Protachykinin-1 is synthesized with a signal peptide (aa 1-19) and a propeptide (aa 20-56), which are subsequently cleaved off. The resulting peptide can be further cleaved into five different peptides- Substance P (aa 58-68), neuropeptide K (aa 72-107), neurokinin A (aa 98-107), neuropeptide gamma (aa 72-107) and C-terminal-flanking peptide (aa 111-126). Substance P is a potent hypotensive peptide that is most well-studied of the tachykinin family of peptides. Its actions are mediated via binding to NK1 receptors. Substance P is widely distributed in the central and peripheral nervous system of vertebrates. It is also found in association with nerves intrinsic to the gastrointestinal tract arising from the submucous and myenteric plexuses. In the CNS, it participates in various behavioral responses and in regulating neuronal survival and degeneration. It also regulates cardiovascular and respiratory function and is involved in activating the emetic reflex. In the spinal cord, substance P participates in neurotransmission of pain and noxious stimuli and modulates autonomic reflexes. It is also reported to elicit local vasodilatation and alter vascular permeability, which enhances the delivery and accumulation of leukocytes to tissues for the expression of local immune responses. It is also known to stimulate the chemotaxis of lymphocytes, monocytes, neutrophils, and fibroblasts. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
