Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GPR146 in human liver and 1:10,000 dilution in rat liver tissue lysates.

Affinity Binding Assay: A representative lot of this antibody bound GPR146 with a KD of 8.0 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GPR146 in LoVo cells.

General description

Probable G-protein coupled receptor 146 (UniProt: Q96CH1; also known as G-protein coupled receptor PGR8, GPR146) is encoded by the GPR146 (also known as PGR8) gene (Gene ID: 115330) in human. GPR146 is a multi-pass membrane protein with four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. It is reported to promote the activity of hepatic sterol regulatory element binding protein 2 (SREBP2) via activation of the ERK signaling pathway that regulates hepatic very low-density lipoprotein (VLDL) secretion, and subsequently reduces the circulating levels of low-density lipoprotein cholesterol (LDL-C) and triglycerides (TG) levels. Deficiency of GPR146 is shown to reduce hepatic SREBP2 activity and VLDL secretion rate. This significantly reduces plasma cholesterol levels in both wild-type and LDL receptor-deficient mice. Mice deficient in GPR146 are shown to have significantly reduced aortic atherosclerotic lesions. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
