Description

Background information

Protein disulfide-isomerase, also known as PDI, is an abundant component of the ER that catalyzes disulfide bond formation in the biosynthesis of various secretory and cell surface proteins. It is a multifunctional polypeptide identical to the β-subunit of prolyl 4-hydroxylase, a α2β2 tetramer that catalyzes the formation of 4-hydroxyproline in collagen by the hydroxylation of proline residues in peptide linkages. PDI has two thioredoxin-like regions, each containing a Cys-Gly-His-Cys sequence that may represent two independently acting catalytic sites of PDI activity. The C-terminus of PDI has a KDEL sequence, which acts as retention signal for proteins of the ER lumen. The PDI/β-subunit of prolyl-4-hydroxylase is required for its retention in the ER.

Physical form

Purified mouse monoclonal IgG1κ in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.

Application

Immunocytochemistry Analysis: A representative lot was used by an independent laboratory in methanol-fixed NRK cells (Vaux, D., et al. (1990). Nature. 345(7):495-502).

Immunogen

Linear peptide corresponding to human Protein disulfide-isomerase.

Quality Assurance

Evaluated by Western Blot in human adrenal gland tissue lysate.

Western Blot Analysis: 0.5 µg/mL of this antibody detected Protein disulfide-isomerase in 10 µg of human adrenal gland tissue lysate.
