General description

SIGMAFAST Protease Inhibitor Cocktail Tablets, EDTA-Free is a tableted formulation containing water-soluble protease inhibitors with broad specificity for the inhibition of serine, cysteine, aspartic and metalloproteases. The tablets have been formulated to be compatible with sample processing by immobilized metal affinity chromatography (IMAC) such as used for the purification of histidine-tagged proteins. The tablets contain no strong metal chelators, such as EDTA, which might interfere with IMAC processing. Finally, the tablets contain a unique, proprietary formulation including a pepstatin A salt to provide additional protection not normally obtained in a tablet format.


Inhibits serine, cysteine, aspartic, and metalloproteases.


The broad protection afforded allows SIGMAFAST Protease Inhibitor Cocktail Tablets to be used for the purification of histidine-tagged proteins, as a general-purpose protease inhibitor cocktail and as components of buffers for cell lysis. The tablets have also been optimized and tested for use with HIS-Select media.


Pepstatin A


One tablet makes 100 mL of cocktail. One tablet is recommended for the inhibition of the proteases present in a maximum of 20 g of cell mass in 100 mL of extract.

Storage and Stability

The tablets are stable as supplied for at least 2 years at 2–8 °C. The reconstituted protease inhibitor solution (1´ or 10´) is stable for at least 2 weeks at 2–8 °C.

What is the difference between Protease Inhibitors P8849 and S8830?
P8849 is a DMSO solution and S8830 is a water soluble tablet. P8849 formulation is considered proprietary but does contain AEBSF, Pepstatin A, Bestatin, E-64, and Phosphoramidon. The formulation of S8830 also contains Aprotinin and Pepstatin A and the exact formulation is given on the product information sheet (under Documents, above).
Can Product No. P8849, Protease Inhibitor Cocktail, be used with with HIS-Select products or any other immobilized metal affinity chromatography (IMAC) products?
This product does not contain EDTA and can be used with sample processing by immobilized metal affinity chromatography (IMAC) suchas used for the purification of histidine-tagged proteins..
How should I dissolve Product No. S8830, SIGMAFAST Protease Inhibitor?
You can dissolve the product in 100 ml of water to make a 1X solution. You can make a 10X stock solution using 10 ml of water and then dilute 1:10 for use.Concentrations greater than 1X may appear slightly hazy. This will not affect the performance of the protease inhibitors.
