Description

Immunogen

synthetic peptide corresponding to amino acids 347-360 of human DNAse II precursor.

Application

Anti-Deoxyribonuclease II (DNase II) antibody produced in rabbit is suitable for microarray and immunoblotting at a working concentration of 0.5-1 μg/mL (1:1000 – 1:500 dilution) using a human spleen lysate. It was used at a working concentration of 0.25 μg/mL to examine the immunoreactivity of samples prepared from outer hair cells of guinea pigs’ inner ear in a study.

Biochem/physiol Actions

DNase II digests DNA at low pH and acts as a lysosomal enzyme responsible for degrading the exogenous DNA that enters the cell. In the nucleus, it degrades the DNA during apoptosis. It causes both chromatin condensation and DNA fragmentation. It is involved in DNA fragmentation induced by lovastatin through activation of caspase-3 and DNase II activities in HL-60 cells. The gene encodes a 40 kDa proenzyme. The mature enzyme consists of two non-identical subunits, the 32 kDa (α) and 12 kDa (β) chains, generated by proteolytic processing.
