Biochem/physiol Actions

Amphiregulin (AREG) is a protein encoded by the AREG gene in humans and is localized to chromosomal region 4q13-4q21, a common breakpoint for acute lymphoblastic leukemia. It is a bifunctional growth modulator that interacts with the EGF/TGF-α receptor to promote the growth of normal epithelial cells and inhibits the growth of certain aggressive carcinoma cell lines. The gene is related to epidermal growth factor (EGF) and transforming growth factor α (TGF-α). AREG is a widely expressed transmembrane tyrosine kinase and is synthesized as a membrane-anchored precursor protein that can engage in juxtacrine signaling on adjacent cells. It plays a critical role in the development and maturation of mammary glands, bone tissue and oocytes. Increased expression of the gene is associated with different pathological conditions (mostly of inflammatory and/or neoplastic nature). AREG is useful in primary colorectal tumor as an indicator of prognosis and as a predictor of recurrence.
