Description

General description

CERK (ceramide kinase) is a Ca2+-dependent lipid kinase. It was first isolated from synaptic vesicles and HL-60 human leukemia cell line. It has high sequence homology to sphingosine kinase type 1, and its catalytic domain shows similarity to that of diacylglycerol kinase. Its pleckstrin homology (PH/PX) domain, found in the N-terminal is responsible for both its activity and localization to cell membrane. Its C-terminal contains a Ca2+/calmodulin domain. It is localized to plasma membrane in association with caveolin and/or lipid rafts.

Biochem/physiol Actions

CERK (ceramide kinase) is responsible for catalyzing the synthesis of the sphingolipid ceramide-1-phosphate (C1P), by phosphorylating ceramide. Ceramide on the other hand, is stimulated by pro-inflammatory compounds to produce prostaglandins. It is involved in Ca2+ signaling, and promotes phagolysosome formation. It is responsible for the phagocytosis of polymorphonuclear leukocytes, and is involved in the degranulation of mast cells. It might also play a role in the differentiation of leukocytes. It might play a role in trafficking of vesicles and exocytosis, as it is localized to trans-Golgi compartments, and possibly to endosomes. It is involved in cell survival, by striking a balance between C1P, which is anti-apoptotic, and ceramide, which is pro-apoptotic.
