Description

General description

ACTR2 (ARP2/actin-related protein 2) is a member of the ARP family, and is localized to the cytoplasm. It has a lipid-binding domain, which facilitates membrane attachment. It is one of the three major classes of ARP family, the other two being ARP1 and ARP3. It is ubiquitous, and has a protein folding pattern structure similar to that of actin. It forms a component of the seven subunit protein complex called Arp2/3.

Biochem/physiol Actions

ACTR2 (ARP2/actin-related protein 2) is a part of the Arp2/3 complex, which is essential for the assembly of actin filaments. Filament assembly is essential for processes such as, endocytosis and cell locomotion. The phosphorylation of Arp2, at threonine or tyrosine residues is essential for the activation of Arp2/3 complex. Abnormalities in the function of Arp2/3 complex result in multiple disorders such as, cancer metastasis. This protein interacts with coronin3, and these proteins together control the invasiveness and metastasis of gastric cancer (GC). Thus, the combination of these two proteins can have potential as a prognostic marker for GC. Inactivation of Arp2/3 complex influences chemotactic signaling non-autonomously, in an NF-κB-dependent manner. In lung adenocarcinoma, the co-expression of Wiskott-Aldrich syndrome family verproline-homologous protein 2 (WAVE2) and ACTR2 is associated with cancer metastasis and poor patient prognosis.
