LONG® R3 IGF-I is a recombinant analog of human insulin-like growth factor-I (IGF-I) that has been specifically engineered for the enhancement of cell culture performance. It is more biologically potent in vitro than either insulin or native IGF-I and has been shown to significantly increase recombinant protein production. It is ideal for both research and large-scale culture systems utilizing serum-free or low-level serum applications. LONG R3 IGF-I has been used with numerous cell types including CHO, vaccine-producing cell lines, fibroblasts and stem cells. All cell types that have a growth response to insulin in cell culture have the potential to respond to LONG R3 IGF-I. LONG R3 IGF-I provides equivalent or better performance to recombinant insulin depending on the cell line and clone.

LONG R3 IGF-I is produced in an Escherichia coli expression system without the use of animal-derived components.

LONG® R3 IGF-I human has been used:
• for the in vitro direct stimulation of pancreatic islets
• as a component of growth medium for retinal cell lines
• to culture vascular smooth muscle cells (VSMCs)

How do the sequences of Products I1146 and I1271 compare to that of native Insulin-like Growth Factor 1 (IGF-1)?
The entire IGF-1 gene codes for 194 amino acids, but the mature form of the protein is 70 amino acids, consisting of amino acids 49-118.Product I1146 is R3 IGF-1, which is also 70 amino acids in length. The Glutamic acid (E) at position 3 has simply been replaced by an Arginine (R). So the sequence of Product I1146 is:GP RTLCGAELVD ALQFVCGDRG FYFNKPTGYG SSSRRAPQTG IVDECCFRSC DLRRLEMYCA PLKPAKSAProduct I1271 is the same as I1146, but with an additional thirteen amino acids "in front of" the normal N-terminus of the R3 IGF-1:MFPAM PLSSLFVNGP RTLCGAELVD ALQFVCGDRG FYFNKPTGYG SSSRRAPQTG IVDECCFRSC DLRRLEMYCA PLKPAKSANote that the first 13 amino acids in the recombinant I1271 (MFPAMPLSSLFVN)are quite different from what would have been the preceding 13 amino acids in the immature IGF-1.
