Application

Determination of the subclass of a monoclonal antibody is helpful for characterization of the antibody, for choosing detection reagents, and for deciding on a purification scheme.

The IsoQuick strips, whether purchased in kits (ISOQ5 and ISOQ25) or separately (I9285, I9410, or I9535), are designed to detect and identify mouse isotypes in cell culture supernatant, ascites, or purified antibody. Optimum detection range is 0.1-4 μg/mL with a detection limit of 10 ng/mL. IsoQuick strips have been validated for use with mouse monoclonal antibodies and have not been validated for use with other host antibodies.

The IsoQuick IgA/IgM strips are designed to detect and identify mouse IgA and IgM isotype subclasses.

Can I useIsoQuick strips to classify the antibody light chains?
Yes, Product No. I9535 is for distinguishing kappa and lambda light chains.These strips can be ordered separately.They are provided in the ISOQ5 and ISOQ25 kits.
With what types of samples willIsoQuick strips work?
The IsoQuick strips can be used in cell culture supernatant with no dilution or other preparation. If testing ascites the sample should be diluted in a range from 1:10,000 to 1:100,000. If testing purified antibody, the sample should be diluted to a concentration of 0.1 to 4.0 μg/mL.
How sensitive is IsoQuick ?
The sensitivity is quite good; each IsoQuick strip can detect as little as 0.1 μg/mL
CanIsoQuickstrips be reused?
No. The strips can only be used once, even if all the test lines did not develop.
Are the IsoQuick strips light sensitive?
Yes. The strips should be kept in the desiccant tube they were delivered in to protect them from light contamination. Prolonged exposure to UV or other light sources may affect the visibility of the control and test lines on the device.
Are the IsoQuick strips humidity sensitive?
Yes. The strips should be kept in the desiccant tube they were delivered in to ensure the proper humidity level. Strips left outside the tube in a high humidity environment may result in test errors.
Are the IsoQuickstrips heat sensitive?
Yes. The strips should be kept in the desiccant tube and stored at 2-8°C. This will ensure maximum useful life. IsoQuick strips can be stored at room temperature, but this will diminish their useful life. If the strips are exposed to extreme hot or cold environments the performance of the strip is likely to be effected.
When using IsoQuick strips, do I need to dilute my samples?
No dilution is required when using the IsoQuick in cell culture supernatant. Place strips directly into the sample. To use IsoQuick strips in ascites dilute the sample to a concentration of 1:10,000 to 1:100,000. To use IsoQuick strips to test purified antibody dilute the sample to a concentration of0.1 - 4.0 μg/mL.
Can I use the IIsoQuickstrips to determine the antibody titer?
IsoQuick IgG strips can be used for semi-quantitative determination of the IgG titer in a cell supernatant sample. For semi-quantitative measurement, run IsoQuick strips with standards of known titer (1,000 μg/mL, 100 μg/mL, and 10 μg/mL). Then compare the color of the result line for the sample to those obtained with the standards (visually or using a color absorbance reader). Color intensity is representative of sample titer.
