Biochem/physiol Actions

JF5 is an allosteric inhibitor of PAR1. The molecule binds to helix 8 of the receptor, directly interfering with receptor coupling to Gαq, but not Gα12. JF5 also inhibits α2A-adrenergic receptor and mPAR4 activity, but not hPAR4. JF5 blocks granule secretion from platelets activated with the PAR1 agonist SFLLRN (IC50 4 μM), but does not affect platelet activation by convulxin (GPVI agonist), PMA or A23187.
