Thioureas

Thioureas are organosulfur compounds structurally similar to ureas, except the oxygen atom is replaced by a sulfur atom to constitute the S=C(NH2)2 moiety. Jacobsen‘s research group has developed a range of chiral thioureas that are versatile and effective organocatalysts. A range of latent nucleophiles can be added to mostly imine-type electrophiles in excellent enantiomeric excesses and, in general, with a broad substrate scope. Thiourea derivatives represent a promising class of anticancer agents because of their inhibitory activity against protein tyrosine kinases (PTKs), human sirtuin type proteins (SIRT1 and SIRT2), topoisomerase II8 and DNA repair synthesis. These enzymatic inhibitory activities coupled with the antiviral, antimalarial, antibacterial and antiinflammatory activities of thioureas have therefore attracted considerable pharmaceutical interest in thiourea derivatives as an important pharmocophore in synthetic medicinal chemistry.
