Understanding biochemical mechanisms, such as signaling pathways is critical for oncology research. These pathways are often evaluated through studying protein-protein interactions and post translational modifications (PTMs). Duolink technology, based on the Proximity Ligation Assay (PLA) is a powerful investigative tool for the detection and visualization of individual proteins, protein-protein interactions and post-translational modifications in fixed samples. The 1000X signal amplification allows for single molecule sensitivity and detection at endogenous levels using regular fluorescent or optical microscopy techniques. Duolink provides additional information to current biomarker expression profiling assays due to its ability to detect and measure functional protein complexes. The activation status of a given signaling pathway, such as receptor tyrosine kinases can be assessed in established cell lines as well as patient biopsies, and xenographs. With circulating or secreted biomarkers Duolink’s dual antibody targeting feature allows for increased specificity (benign versus malignant), sensitivity and novel biomarker identification. The ability to evaluate and quantify these proteins within a native state allows researchers to better understand the complex mechanisms that both develop and are a result of tumorigenic conditions.
