3D Scaffolds

Tools allowing the growth of cells in a 3-dimensional environment are generally divided into different approaches, including so called scaffold-free and scaffold-based technologies.
3D Scaffolds, traditionally described as tools made of polymeric biomaterials, have the advantage to provide a structural support for cell attachment and tissue development. Scaffolds allow recapitulation of the extracellular environment of cells, the ECM (Extracellular Matrix), by providing attachment sites, the ability for cells to grow in 3D shape, and for some of them, rigidity of this environment and associated soluble factors (growth factors, immune system).
Numerous scaffolds have been developed by innovative companies around the world, and at MilliporeSigma we have chosen to offer a panel of these technologies to the scientific community.

Collagen Scaffolds

SpongeCol® is a collagen sponge with an interpenetrating, columnar porous architecture structure. It contains a unique columnar porous network which permits cells and nutrients to flow completely through the interpenetrating pores and provides an increased surface area for cell attachment, growth and migration. SpongeCol® is composed of highly purified Type I collagen which best supports the attachment, proliferation, and function of cells.
SphereCol®, human collagen coated beads is coated with highly purified Type I human collagen derived from a human fibroblast cell culture process, VitroCol® . The collagen provides an optimal coating on the beads to enhance cell attachment, cell viability, cell proliferation and cell function.

Gelatin Scaffold: Go Matrix gelatin based scaffolds

Go Matrix is a gelatin based rigid material enabling easy manipulation (presented as a dry insert). Because the pore are uniforms (pore size is 60, 90 or 130 μm), it offer a uniform micro-environment. This open network allows efficient cells communication.
Go Matrix has been tested with a variety of cells (MDCK cells, C2C12 Myoblast, 3T3 Fibroblast, Cardiomyocyte, CL1-0, CL1-5 & A549…).

Lena Biosciences 3D Cell Culture Systems

GradientEZ is a versatile system for 3D cell migration assays. It is exceptionally easy to use, suitable for generating gradients of test compounds, and testing of drug combination strategies with adherent or suspension cells in situ. Simply, spot-a-culture, a drug, or an extracellular matrix. Alternatively, stack the substrates to generate gradients. GradientEZ enables parallel testing of plurality of drugs, drug gradients, or drugs in different concentrations.
SeedEZ scaffold is a complete 3D cell culture and drug screening system; a convenient 3D framework for 3D cell-based assays and organ-like tissue models. It accepts most cell types, hydrogels and extracellular matrices, active and inactive biologicals, polymers for drug delivery and controlled drug release, drug excipient formulations, and test compounds.

Nanofiber Scaffolds

Nanofiber cell culture dishes, multiwell plates and well plate inserts combine the best of 3D and 2D cell culture. The nanofibers are optically transparent to allow for live-cell imaging and real time quantification of cell mobility. The nanofibers mimic the 3D topography in vivo, and can be easily coated with ECM proteins for Cancer Research. The Cells can be easily removed for protein or gene analysis.

The nanofibers have high expansion rates, excellent differentiation control, and synthetic surfaces with no animal contamination; they are ideal for stem cell research and immunohistochemistry staining. The standard plate dimensions are compatible with current automated equipment for high throughput drug discovery needs. The cell culture dishes, plates and inserts are available with either randomly oriented fibers or aligned fibers.

Polystyrene Scaffolds

Polystyrene (PS) is the polymer that is used in the fabrication of various cell culture vessels, such as tissue culture plates (TCP) and tissue culture flasks. They are non-cytotoxic and crystal clear. Similarly, PS 3D Insert scaffolds are tissue culture surface treated and transparent. The transparency of the PS 3D Insert scaffold is a considerable advantage compared to other porous scaffolds on the market because it makes it possible for researchers to directly observe the cell growth under routine cell culture inverted microscopy. Ultimately, this means that 3D cell culture can be conducted very continently and economically without the need of adding expensive instruments. It also means that all routine 2D cell culture protocols can easily be adapted to 3D cell culture with few changes.

Scaffdex Culture Products

CellCrown is a disposable insert, which immobilizes a sample in its place on a 6-, 12-, 24-, 48-, and 96- well plate. Fibers, foils, membranes, textiles, tissue and block like samples can be fixed with CellCrown in a firm, adjusted position in the well. The sample can be left in the well during the change of media by means of a pipette between the wall of the well and the CellCrown.

CellVessel is made of bioabsorbable multifilament (8-filaments) poly-96L/4D-lactide copolymer fiber. Poly-96L/4D-lactide copolymer has been tested with various cell types and it works especially well with fibroblasts.
