Trypsin Substrates

Kinetic measurement of trypsin activity is usually followed spectrophotometrically using arginine esters or amides as substrates. Cleavage of BAEE (Na-Benzoyl-L-arginine-ethyl-ester-hydrochloride) is measured in the ultraviolet range, while arginine nitroanilide and napthylamide substrates produce colored products when cleaved , and thus are useful in colorimetric assays. Several of these chromogenic substrates are tripeptides, altering their specificity. For example, Pefachrome Fxa is primarily used as a substrate for clotting factor Xa. Researchers can also follow amidase and esterase activity flurometrically, with 4-methylcoumarin and methylumbelliferin substrates respectively.
