Traditional Calcium Indicators

Fura 2 is a UV light-excitable, ratiometric Ca2+ indicator that is a common dye of choice for ratio-imaging microscopy. Upon binding Ca2+, Fura 2 exhibits an absorption shift that can be observed by scanning the excitation spectrum between 300 and 400 nm, while monitoring the emission at ~510 nm.

Another ratiometric, UV light-excitable Ca2+ indicator, Indo 1, is the preferred dye for flow cytometry and uses a single laser for excitation. The emission maximum of Indo 1 shifts from ~475 nm in Ca2+-free medium to ~400 nm when the dye is saturated with Ca2+.

Fluo 3 uses the same typical excitation sources used in flow cytometry and confocal laser scanning microscopy as well as drug screening applications. Fluo 3 is essentially non-fluorescent unless bound to Ca2+, but shows approximately a 40-fold increase in fluorescence when bound to Ca2+.

Calcein AM is an outstanding indicator of cell viability. When compared to other cell viability indicators, viable cells retain calcein better and calcein does not interfere with various immunological processes like leukocyte chemotaxis or lymphocyte-target cell conjugation.
