FLAG® Affinity Gels

The FLAG® tag was designed for fusion protein purification via immunoaffinity chromatography. Our FLAG® affinity gels feature anti-FLAG® antibodies (M1 or M2) covalently attached to a variety of solid supports, making these gels ready-to use in your protein purification workflow.

FLAG® fusion proteins are most commonly purified with M1 or M2 anti-FLAG® antibodies by either competitive elution with FLAG® or 3xFLAG peptides or by using a reduced pH buffer. Additionally, as the binding of M1 anti-FLAG® antibodies is calcium-dependent, anti-FLAG® M1 agarose affinity gels can also be used for the calcium-mediated purification N-terminal FLAG® fusion proteins. anti-FLAG® M1 agarose affinity gels cannot be used for the purification of Met-FLAG® fusion proteins.

A4596 ANTI-FLAG® M1 Agarose Affinity Gel ≥0.6 mg/mL, gel binding capacity
For purification of N-terminal FLAG fusion proteins. Since binding is Ca2+-dependent, proteins can be eluted with a buffer containing EDTA, as well as by the standard methods using either FLAG peptide or glycine-HCl buffer, pH 3. ANTI-FLAG M1 does not bind to Met-FLAG fusion proteins, so this resin is not appropriate for purifying unprocessed, cytoplasmically expressed fusion proteins.

Affinity gel is for calcium mediated purification of N-terminal FLAG fusion proteins.
