FLAG® Antibodies

Three monoclonal anti-FLAG® antibodies (M1, M2, and M5) have been produced for the FLAG® tag sequence, each with different epitope binding requirements.

M1
M1 anti-FLAG® antibody binding is calcium-dependent and is specific for the N-terminus of the FLAG® tag. The M1 anti-FLAG® antibody cannot be used with Met-FLAG® (N-terminus preceded by a methionine residue) or C-terminal FLAG® fusion proteins. When used in affinity chromatography, bound antigens can be eluted using chelating agents (e.g. EDTA), in addition to competitive elution or reduced pH buffers.

M2
M2 anti-FLAG® antibodies bind N-terminal FLAG®, C-terminal FLAG®, and Met-FLAG® fusion proteins. The universal nature of this antibody renders it more generally applicable to a wide variety of applications. Bound antigens can be eluted during affinity chromatography via competitive elution or the use of reduced pH buffers.

M5
While M5 anti-FLAG® antibodies can be used with both types of amino-terminal FLAG® fusion proteins, they exhibit a higher affinity for Met-FLAG® fusion proteins. Due to this affinity, M5 anti-FLAG® antibodies are typically used with cytoplasmically expressed FLAG® fusion proteins. Similar to M2 anti-FLAG® antibodies, bound antigens are eluted by competitive elution with FLAG® peptides or reduced pH buffers.

F4042 Monoclonal ANTI-FLAG® M5 antibody produced in mouse using Protein A
Binds the FLAG peptide only when it is located at the amino terminus preceded by a methionine. Binding is not Ca2+-dependent. Useful for detecting cytoplasmically expressed Met-FLAG fusion proteins in mammalian crude cell extracts, but not recommended for fusion proteins expressed in E. coli.
Suitable for western blotting.
