FLAG® Affinity Purification

The FLAG® peptide, Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys, was devised for use as an epitope tag in fusion proteins1. It can be linked to either the carboxyl-terminus or amino-terminus and can be easily cleaved away by enterokinase. However, because the FLAG® peptide is so hydrophilic, it normally does not interfere with protein folding or function, and enables purification in non-denaturing conditions. FLAG®-tagged proteins have been used extensively in plant protein research, especially in Arabidopsis and tobacco. For affinity purification of these recombinant proteins, our M2 monoclonal antibody to FLAG® is bound to agarose; target protein can be eluted with FLAG peptide or with 3X FLAG®. Our convenient FLAG® immunoprecipitation kit provides buffers and a positive control fusion protein in addition to the affinity matrix and 3X FLAG®. EZview red anti-FLAG® M2 affinity gel greatly simplifies immunoprecipitation of FLAG® proteins because it is easy to detect accidental aspiration of the red agarose. M2 monoclonal is also available bound to a 96-well microtiter plate via its Fc region for sensitve, high through-put screening of fusion proteins.
