Analysis Note

These secondary standards offer multi-traceability to the USP and EP (PhEur) primary standards, where they are available.

Application

Primarily used as an antibacterial agent. Dihydrofolate reductase inhibitor with selectivity for the prokaryote enzyme.
These Secondary Standards are qualified as Certified Reference Materials. These are suitable for use in several analytical applications including but not limited to pharma release testing, pharma method development for qualitative and quantitative analyses, food and beverage quality control testing, and other calibration requirements.
Trimethoprim may be used as a pharmaceutical reference standard for the determination of the analyte in pharmaceutical formulations by chromatography[1][2] and visible and UV spectrophotometry.[3]

General description

Certified pharmaceutical secondary standards for application in quality control provide pharma laboratories and manufacturers with a convenient and cost-effective alternative to in-house working standards. Trimethoprim is a synthetic antibacterial agent active in vitro against most aerobic gram-negative and gram-positive bacteria. Its mode of action involves inhibition of dihydrofolate reductase, an enzyme involved in catalyzing the reduction of dihydrofolate to tetrahydrofolate in both microbial and eukaryotic cells.[4]

Questions

1–5 of 5 Questions
  1. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. In what is Trimethoprim best soluble ? Can it be dissolved in HCl?

    1 answer
    1. Trimethoprim is soluble in ethanol at 50 mg/mL, in water at 0.4 mg/mL, in DMSO at 50 mg/mL, in dimethylacetamide at 138.6 mg/mL, in benzyl alcohol at 72.9 mg/mL, in propyleneglycol at 25.7 mg/mL, in chloroform at 18.2 mg/mL, and in methanol at 12.1 mg/mL at room temperature. The solubility in HCl is not determined.

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  5. Is Trimethoprim stable in solution?

    1 answer
    1. Triemethoprim in a solution of 52% N,N dimethylacetamide:48% propylene glycol at 50 mg/ml is stable when stored at 25° C. There is only a 10% degradation of trimethoprim under these conditions after 885 days.
