Allosteric site-targeting macrophage migration inhibitory factor (MIF) inhibitor that protects HeLa cells against MNNG-induced parthanatos.
MKA031 is an allosteric site-targeting macrophage migration inhibitory factor inhibitor (MIF tautomerase activity IC50 = 1.7 μM) that prevents MNNG-induced MIF nuclear translocation (3h 100 μM MKA031 preincubation prior to 15-min 50 μM MNNG treatment; HeLa cells) by interfering with MIF/AIF interaction (IC50 = 12.3 μM against 4 μM AIF in wells precoated with 100 μL of a 250 nM MIF solution). Unlike ZP143, MKA031 protects HeLa cells against MNNG-induced parthanatos (EC50 = 7.7 μM 24h post 15-min 50 μM MNNG treatment).
