Application

JNJ-10397049 may be used in proliferation assay to study its role in growth and differentiation of neural progenitor cells (NPCs).

Biochem/physiol Actions

JNJ-10397049 is a potent and subtype-selective orexin receptor 2 (OX2R) antagonist (pKi = 8.3/OX2R vs. 5.5/OX1R) with little affinity toward a panel of 50 receptors, ion channels and transporters. Blockade of OX2R by JNJ-10397049 antagonizes against orexin B-induced cellular calcium response (pKB = 7.9) and attenuates ethanol self-administration, place preference, and reinstatement in rats in vivo (1, 3, and 10 mg/kg, sc).
