Biochem/physiol Actions

Nimustine HCl (ACNU) is a brain-penetrating nitrosourea that kills cancer cells by damaging their DNA. Nimustine HCl forms O6-chloroethylguanine adducts, which cross-link with other DNA strands, blocking replication and leading to cell death. Nimustine also suppresses blood cell production and is often used in combination with other chemotherapy drugs. Nimustine undergoes conversion into several active metabolites, including denitrosated ACNU, 3-((4-amino-2-methylpyrimidin-5-yl)methyl)-4-hydroxy-1-nitrosoimidazolidin-2-one, 1-(2-chloroethyl)-1-nitrosourea and 1-((4-amino-2-methylpyrimidin-5-yl)methyl)-5-hydroxyimidazolidin-2-one.
