Description

Analysis Note Sample preparation: The dust of a vacuum cleaner was sieved through a normal kitchen sieve. 500 mg of the sieved dust was suspended with 5.4 mL potassium hydroxide (10%) in methanol and stirred over 2h by 80°C.After cooling down, the suspension  was centrifuged. 4mL of the obtained solution was filtered through a 0.45 μm  membrane filter and washed with 2mL water. The combined solutions were then extracted two times with 2 mL n-hexane. The n-hexane was evaporated with nitrogen gas and the residue was resolved in 1 mL methanol. The  final solution  were  spiked with 100 μL of Ergosterol  (1 mg/mL) standard solution .
