RNAi Transfection

The goal of this exercise is to learn the importance of testing different concentrations of RNAi reagents to achieve optimal knockdown efficiency of the targeted gene, and observe phenotypic changes in the cells 24 hrs post-transfection. or this example we will be using a Chinese hamster ovary cell line CHO-K1 that has previously been engineered to exhibit stable TurboGFP expression. In this exercise we will transfect these cells with the si, esi, and shRNAs targeting TurboGFP with the goal of knocking down GFP expression.
