Application

Aprotinin wird im Wesentlichen als Trypsin-Inhibitor eingesetzt.

Biochem/physiol Actions

Aprotinin ist ein kompetitiver Inhibitor der Serinprotease, der stabile Komplexe mit Enzymen bildet und aktive Enzymstellen blockiert. Diese Bindungen sind reversibel und die meisten Aprotinin-Protease-Komplexe lösen sich bei extremen pH-Werten von >10 oder <3 auf. Strukturell ist Aprotinin ein monomeres globuläres Protein; ein Derivat aus der Rinderlunge, das aus 58 Aminosäuren besteht, die in einer einzelnen Polypeptidkette mit drei vernetzenden Disulfidbrücken angeordnet sind.

Eine Trypsin-Inhibitor-Einheit (TIU) senkt die Aktivität von 2 Trypsin-Einheiten um 50 %, wobei eine Trypsin-Einheit bei pH 7,8 und 25 °C 1,0 μmol N-α-Benzoyl-DL-arginin-p-nitroanilid (BAPNA) pro Minute hydrolisiert.

Eine Trypsin-Inhibitor-Einheit (TIU) senkt die Aktivität von zwei Trypsin-Einheiten um 50 %, wobei eine Trypsin-Einheit bei pH 7,8 und 25 °C 1,0 μmol N-α-Benzoyl-DL-arginin-p-nitroanilid pro Minute hydrolisiert. Eine weitere häufig verwendete Einheit ist KIU, wobei 1 TIU = 1.300 KIU.
