General description
Eine hochspezifische Amplifikation ist für eine erfolgreiche qPCR mit SYBR Green I-Färbetechnologie unerlässlich, da dieser Farbstoff an alle während der Amplifikation erzeugten dsDNA-Moleküle bindet und diese nachweist. Hot-Start-Taq-DNA-Polymerase ist Antikörper-vermittelt und somit vor dem anfänglichen PCR-Denaturierungsschritt inaktiv.
Application
Unterschiedliche Echtzeit-PCR-Systeme verwenden unterschiedliche Strategien für die Normalisierung von Fluoreszenzsignalen und eine Korrektur von optischen Well-zu-Well Variationen. Es ist entscheidend, das für Ihr spezifisches Instrument geeignete qPCR-Reagens zu wählen. KiCqStart SYBR Green qPCR ReadyMix, iQ enthält Fluoreszein für die experimentelle Sammlung von Platten-Well-Faktoren auf den iCycler iQ-Echtzeit-Erkennungssystemen oder auf dem MyiQ-Erkennungssystem.

KiCqStart SYBR Green qPCR-Fertigmix ist bei lichtgeschützter Lagerung in einem Gefrierschrank mit konstanter Temperatur bei -20 °C 1 Jahr lang stabil. Zur bequemeren Nutzung kann der Mix ungefroren bis zu 6 Monate bei +2 bis +8 °C aufbewahrt werden. Nach dem Auftauen die Bestandteile vor der Verwendung gründlich mischen. Wiederholtes Einfrieren und Auftauen wird nicht empfohlen. Nichtsdestotrotz zeigte das Produkt nach 20 Einfrier-/Auftauzyklen bzw. 2 Monaten Lagerung bei +20 °C keinen Leistungsverlust.
