Application

New methylene blue N has been used as a photosensitizer for the photodynamic inactivation of Candida albicans.[1][2]

Biochem/physiol Actions

New methylene blue N can bind, neutralize and crosslink with RNA. This causes co-precipitation of ribosomal and residual RNA with the mitochondria and ferritin masses. These events make the living young erythrocytes as microscopically visible, dark-blue clusters and filaments, referred to as reticulum. New Methylene Blue N helps in counting of reticulocytes (erythrocytes containing RNA).[3]

  1. I would like to use this to stain blood smears to identify/count reticulocytes. What volume of stain would 1g of the powder make?

    1. Instructions for use are not available for this product. However, product R4132 is a prepared solution specifically for use in reticulocyte staining. The New Methylene Blue concentration of this solution is 1% or 1g/100 mL The product formulation is as follows:
      New Methylene blue, 1%
      Sodium Chloride, 0.9%
      Potassium Oxalate, 1.6%

      Based on the formulation listed above, the end-user will be able to prepare 100 ml of a 1% solution if using product 556416. This would also require the addition of the sodium chloride and potassium oxalate components.
