General description

Fibronectin 1 is a glycoprotein of the extracellular matrix that is coded by FN1 gene. It is expressed in the plasma and at the cell surface. The FN1 gene mapped to human chromosome 2q35.[1]
FN occurs as a dimeric protein, linked by two disulfide bonds at the end of C-terminal.[2] Within the fibronectin structure, the domains are compactly arranged close to both the amino and carboxyl terminal of the peptide chains.[3] Moreover, it has been observed that despite arising from a single gene up to 20 distinct variants of this gene have been identified in humans.[2]

Application

Fibronectin from human plasma has been used in the determination of H2O2 consumption rate.[4] It has also been used in coating the Sylgard® models for cell culture.[5] It has also been used in cell adhesion and to directly pattern the protein onto the polymeric film.[6][7]

Biochem/physiol Actions

Fibronectin function encompasses a diverse range of biological processes including cell adhesion, growth, migration, wound healing, blood coagulation and metastasis.[1] Fibronectin protein plays an important role in cell attachment and spreading, control of cell cytoskeleton, morphology and differentiation. FN1 is also involved in extracellular matrix formation, hemostasis and thrombosis. Mutations in FN1 results in glomerulopathy.[8]

Preparation Note

This product is a 0.1% solution of fibronectin. The protein homogeneity is evaluated by immunoelectrophoresis, is endotoxin tested, and the source material was tested negative for HIV, hepatitis B and hepatitis C.

In coating culture surfaces, fibronectin should be diluted in sterile balanced sterile salt solution and coated with minimal volume. Surface should then be air dried for 45 minutes at room temp and can be stored for 2-4 weeks at 2-8°C

Analysis Note

Protein homogeneity is evaluated by immunoelectrophoresis.

    1. The recommended storage duration for Fibronectin (F0895) is up to 1 year at 4°C. There has been no testing done on its stability after storage at -80°C or -20°C. A diluted solution of 10ug/mL in PBS can be stored at 4°C for 2-4 weeks. However, the solution cannot be reused to coat culture ware surfaces multiple times; the recommended process involves coating surfaces with a minimal volume, allowing them to air dry for 45 minutes at room temperature, and then storing the cultureware at 4°C for 2-4 weeks.

    Why do you not have a protocol for using this expensive reagent?

    You have no protocol online for using this very expensive item......why? I have been waiting to chat for a total of 15 minutes to get what you should have put online anyway

    No issues, seems to work for hiPSC differentiation protocols.
