General description

In humans, type I collagen is the most abundant and broadly expressed collagen. This heterotrimer contains two alpha 1 (I) chains and one alpha 2 (I) chain.[1] Collagen type I α 1 (COL1A1), a major component of collagen type I, codes for pro-α 1 chains of type I collagen.[2] It has repeating Gly-Xaa-Yaa tri-peptides motifs. The collagen alpha-1(I) chain protein is a member of the collagen complex superfamily.[1] COL1A1 has a von Willebrand factor type C (vWFC) domain, a collagen triple-helix repeat, and a fibrillar collagen C-terminal domain (COLF).[2]

Application

Collagen Type I, rat tail has been used:
  • for collagen 3D proliferation assays[1]
  • for atelocollagen proliferation assays[1]
  • in a comparative study to make microcapsules for templating on mesoporous cores of pure vaterite calcium carbonate (CaCO3) crystals to evaluate the structure-property relationship[2]
  • in coverslip coating to grow the human cerebral microvascular endothelial cell line (hCMEC/D3) for creating a cell coculture and chronic subdural hematoma (CSDH) patient hematoma sample induced injury model[3]

Biochem/physiol Actions

Collagen type I α 1 (COL1A1) plays a key role in chemoresistance and may act as a therapeutic target. It is involved in carboplatin-resistant ovarian cancer (OV).[1] COL1A1 serves as an oncogene in the development of colorectal cancer (CRC).[2] Dominant inherited COL1A1 gene mutations result in connective tissue disorders such as osteogenesis imperfecta (OI).[1]

    1. The gelation process for this product does not require pH neutralization by the addition of acid or base. The procedure involves pipetting undiluted collagen into culture dishes, exposing them to ammonia vapor, and rinsing with sterile distilled water multiple times to remove ammonia. The final rinse is replaced with a sterile medium or balanced salt solution with serum, and the dishes are stored at 4°C until use.

  3. Accidentally, I opened the collagen out of the flow chamber. Should I filter it or autoclave it to ensure the sterile conditions?

    1. Collagen solutions may be sterilized by dialysis in a 0.25% acetic acid and 0.5% chloroform solution.
      Avoid sterilizing the collagen solution by membrane filtration, as substantial protein loss may occur by this method. The coating of tissue culture plastic dishes may be performed by air-drying. Dried coated dishes may then be sterilized by exposure to UV light in a sterile culture hood or by rinsing with 70% ethanol.

  5. Once diluted in 0.02N acetic acid, does the collagen solution require sterilisation for use in cell culture? If so, which method is the most effective: UV, autoclave or 0.22uM sterile filter (should this be PES or PVDF)?

    1. This product is supplied as a sterile solution in 0.02 N Acetic acid. The package size is 100 mg, however, the concentration is variable and reported on the lot-specific Certificate of Analysis. The typical range is 4 - 5 mg/mL. Dilutions can be made with sterile PBS. Further sterilization is not required. If necessary, coated slides and coverslips can be sterilized by UV.
