General description

Ferritin, an iron-storage protein is usually present in the liver and spleen in mammals. Iron and ferritin are distributed relatively same in the eye. Ferritin is made up of heavy and light chains.

Application

  • Anti-Ferritin, Human antibody produced in rabbit has been used in:
  • immunohistochemistry
  • western blotting
  • immunocytochemical techniques

Anti-Ferritin, Human antibody is suitable for use in Ouchterlony double diffusion (1:16 using human spleen ferritin), and immunoelectrophoresis. The antibody may also be used in western blot (1:1000 using frozen liver homogenates) and immunoprecipitation (using 300 μg of cell lysate incubated with 6 mul of anti-ferritin for 1h at 4 ° C).

Biochem/physiol Actions

Ferritin is an iron-storage protein that regulates iron-homeostasis in cells. While mitochondrial ferritin is a homopolymer, the cytosolic form consists of heteropolymeric L- and H-chains. Genetic mutations in ferritin L-chains have been associated with neurodegenerative disorders, whereas altered levels of ferritin have been linked to hemophagocytic lymphohistiocytosis and adult-onset Still′s disease . In immunoelectrophoresis, anti-ferritin, human antibody is specific for ferritin when used against human spleen extract and purified ferritin. No reactivity is observed against normal human serum.

Preparation Note

The antiserum has been treated to remove lipoproteins.
