Application
SSC Buffer, 20X Concentrate, is a standard reagent in Southern and Northern blotting procedures. This concentrated formula can be used straight or diluted to make SSC working solutions with concentrations as low as 0.5X. It is also for use in preparing nucleic acids for hybridization.
Physical form
This SSC Buffer, 20X Concentrate, contains 0.3 M sodium citrate in 3 M NaCl. Preparation of 1X SSC working buffer: Dilute the 20X concentrated buffer 20-fold with ultrapure water (= 18 MΩ x cm resistivity at 25 °C).
Notes: If precipitation is present in the 20X concentrated buffer, warm the bottle to 37 °C and mix until completely dissolved prior to dilution. It is recommended 1X working solutions be filtered through a 0.2 mm filter before use.
