General description

Cycloheximide solution is a selective supplement, that is safer and easier to use than cycloheximide powder. The required volume can be simply added to the medium before or after sterilization.

Application

Cycloheximide solution has been used as a supplement in De Man, Rogosa and Sharpe (MRS) and GM-17 agar plates to study its effects on lactic acid bacteria (LAB).

Biochem/physiol Actions

Cycloheximide solution is used to block the protein synthesis for suppressing the growth of fungi.

Physical form

Sterile filtered solution containing 0.1% cycloheximide

Questions

1–2 of 2 Questions
  1. Can be used for ribosomal profiling, as a blocker of protein synthesis in eukaryotic cells?

    1 answer
    1. Cycloheximide is commonly used to block protein synthesis in eukaryotic cells and in ribosome profiling experiments. It is recommended to refer to literature for further information, for example: https://pmc.ncbi.nlm.nih.gov/articles/PMC2831214/ and https://www.nature.com/articles/s41467-021-25411-y.

  2. I am looking to add this solution to a Lee's Multi Differential Agar (LMDS) media solution to prevent saccharomyces cerevisiae growth on my plates. How much of this cycloheximide solution should be added per 100 ml of LMDA agar media?

    1 answer
    1. According to brewing industry experts, cycloheximide can be added to Lee's Multi Differential Agar in order to make the media selective to bacteria and wild yeast. With cycloheximide, one can suppress the growth of most brewing yeasts without interfering with bacteria and wild yeast growth. Concentrations recommended for this purpose are in the range from 7-10 mg/L. Adding 10 mLs of product 18079 (at a concentration of 0.1%) to 1 L of agar will result in a concentration of 10 mg/L (a 1:100 dilution).
