General description

Yeast is an inactive edible single-celled organism widely used in the food industry for baking, winemaking, and brewing. This product is dried irrespective of enzyme activity. Yeast serves as a substrate for lyticase.

Application

Yeast has been used:
  • as a structure modifier to alter the morphology and rheology of phase-separated biopolymer gels
  • to induce hyperthermia in rats to perform antipyretic activity test
  • in the extraction and enrichment method for the metaproteomic characterization of dissolved organic matter in groundwater samples

  1. I would like to know how to prepare the product for using it as a substrate in Lyticase activity assay: in what concentration, heat incubation? sonication?. I will appreciate any detailed information. Thank you

    1. The Sigma Enzymatic Assay for Lyticase utilizes Baker’s yeast as the substrate, following the protocol outlined below:

      While brewer’s yeast may be a suitable alternative, it is not guaranteed to exhibit identical behavior. Applying the same protocol is a reasonable starting point; however, experimental validation is strongly recommended to ensure reliable and reproducible results.

      Also, 1U of 04963 Sigma-Aldrich Lyticase from Bacillus subtilis corresponds to the amount of enzyme which decreases the absorbance at 800 nm by 0.001 per minute at pH 7.5 and 25 °C (Yeast, Cat. No. 51475, as substrate).
