Ethylester prodrug of a potent and selective sirtuin 5 (SIRT5) inhibitor against SIRT5-dependent cancer proliferation and survival.
RD167 is the ethylester prodrug of a potent and selective sirtuin 5 (SIRT5) inhibitor (IC50 = 370 nM; <10% inhibition of SIRT1/2/3/6 at 10 μM). NRD167 exhibits anti-proliferation (IC50 = 5-8 μM) and apoptosis-induction efficacy (>80% at 5-8 μM) against SIRT5-dependent, but not SIRT5-independent cells. NRD167 selectively impairs proliferation of AML, but not normal hematological progenitors (6.25-12.5 μM; CD34+ cells). Pretreatment of AML patients-derived cells by NRD167 (12.5-25 μM for 72 hrs) prior to xenograft greatly prevents the survival of injected mice.
