Biochem/physiol Actions

SiR-DBCO is a cell-permeable, silicon-rhodamine strained alkyne click reagent, essential in forming the MAO-SiR complex when used with MAO-N3 for mitochondrial imaging. In studies, this combination showed remarkable stability in HeLa cells, maintaining signal integrity over 12.5 hours with minimal decay, a clear indication of effective mitochondrial labeling. This contrasts sharply with traditional dyes like MitoTracker Deep Red, which often exhibit significant signal loss within hours. Thus, SiR-DBCO, in conjunction with MAO-N3, enhances the duration and quality of mitochondrial imaging, even under lower-intensity lighting conditions. Note: In time-lapse imaging, cells treated with 200 nM MAO-N3 and 100 nM SiR-DBCO, collectively known as MAO-SiR, exhibit minimal signal degradation and a rise in cell density, indicative of robust mitochondrial function and cell health. This outcome is in stark contrast to the results observed with dyes like MitoTracker Deep Red, which typically show a substantial decline in signal intensity within just a few hours.
