General description

Eravacycline, a synthetic fluorocycline antibiotic, belongs to the tetracyclines class. It is structurally similar to tigecycline, featuring two modifications on the D-ring of its tetracycline core: a fluorine atom substitutes the dimethylamine group at position C-7, and a pyrrolidinoacetamido group replaces the 2-tertiary-butyl glycylamido at position C-9.

Application

Eravacycline hydrochloride may be used as a model for studying antibiotic resistance and the development of new antimicrobial agents.

Biochem/physiol Actions

Eravacycline is a tetracycline class antibiotic with broad-spectrum activity against both Gram-positive and Gram-negative bacteria. It inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, preventing the incorporation of amino acid residues into elongating peptide chains. Its structural modifications allow it to maintain activity against bacteria that express ribosomal protection proteins and efflux pumps. Eravacycline is potent against a variety of pathogens, including methicillin-resistant Staphylococcus aureus (MRSA) and carbapenem-resistant Enterobacteriaceae. Eravacycline demonstrates minimum inhibitory concentrations (MICs) that are two to eight times lower than those of tigecycline when tested against carbapenem-resistant Acinetobacter baumannii (CRAB).
