General description

MTSEA biotin-XX is a membrane-impermeant crosslinker consisting of biotin-coupled 2-aminoethyl-methanethiosulfonate (MTSEA).

Application

MTSEA-Biotin-XX may be used to covalently label cysteine residues in proteins or thiol-containing molecules with a biotin tag for streptavidin-based detection, purification, or accessibility studies and, for biotinylation of 4TU‐labeled RNAs in direct RNA sequencing.

Biochem/physiol Actions

MTSEA-Biotin-XX selectively and efficiently labels cell surface thiols, including cysteine residues and specific RNA modifications (s4U and dm6A), forming disulfide bonds with enhanced reactivity. The extended -XX- linker facilitates optimal biotin-streptavidin interaction, while the overall structure allows for high-yield, rapid reactions. Biotin-XX-MTSEA enables imaging, purification, and sequencing applications through its ability to tag target molecules via methanethiosulfonate (MTS) reactions.
