Application

AT2R agonist C21 may be used to study:
  • its vasorelaxant effect of C21 in mouse mesenteric arteries derived from wildtype and AT2R-knockout (AT2R-/y) mice
  • its effects on U46619-induced β-arrestin recruitment to TP-receptors and the effect of C21 on U46619-induced platelet aggregation

Biochem/physiol Actions

C21 (Buloxibutid; Compound 21; M024) is a potent and selective angiotensin II receptor (AT2R) nonpeptide agonist (Ki = 0.4 nM/AT1R vs. >10 µM/AT1R) that induces AT2R-mediated morphological changes in NG 108-15 neuroblastoma-glioma hybrid cells (100 nM for 3 days), blockable by AT2R antagonist PD 123,319 (1 µM). C21 enhances in vivo duodenal alkaline secretion in Sprague-Dawley rats (30 & 300 µg/kg/h via i.v.), and lowers the mean arterial blood pressure in anesthetized, spontaneously hypertensive rats in vivo (50 µg/kg/h via i.v.). C21 shares structural similarities with AT1-receptor antagonists irbesartan and losartan, and may show TP-receptor antagonistic properties.
