General description

BUPY, a novel dibromopyrazinium-based rebridging agent, offers rapid and efficient site-selective modification of antibody interchain disulfide bonds, achieving highly homogeneous conjugates with a degree of labeling of 4 and 97% rebridging efficiency. BUPY’s excellent aqueous stability, fast reactivity, and ability to function in slightly acidic conditions make it advantageous for antibody conjugation. The terminal alkyne handle enables facile click-chemistry functionalization with therapeutic or diagnostic payloads, while preserving the antibody′s target recognition capabilities. BUPY′s versatility and effectiveness position it as a valuable tool for researchers developing next-generation antibody-drug conjugates and diagnostic antibody probes.

Application

Clickable Antibody Rebridger, BUPY may be used to site-selectively reconnect reduced interchain cysteines on IgG antibodies, such as trastuzumab, enabling the creation of homogeneous conjugates that can be further modified with fluorophores or cytotoxic payloads. It may also be used in in the dSEC-MS workflow to quantify the efficiency and quality of rebridged antibodies, enabling real-time monitoring of antibody rebridging, assessment of critical quality attributes, and verification of full rebridging versus half-antibody species in BUPY-modified monoclonal antibodies.
